Selection and Optimization of Medium Components for the Efficient Production of L-Asparaginase by Leucosporidium scottii L115—A Psychrotolerant Yeast

This study reports the production of L-asparaginase (ASNase), an enzyme mainly used for treatment acute lymphoblastic leukemia, by Leucosporidiumscottii L115, a psychrotolerant yeast isolated from Antarctic ecosystem. Focus was given to select most appropriate medium components able maximize this yeast, as first step development new process produce ASNase. By combining knowledge in bioprocesses, statistical analysis and modeling, composition that favored established, which consisted using mixture sucrose (28.34 g L?1) glycerol (15.61 carbon sources, supplemented with proline (6.15 following salts (g L?1): KCl, 0.52; MgSO4·7H2O, CuNO3·3H2O, 0.001; ZnSO4·7H2O, FeSO4·7H2O, 0.001. medium, 2850 U L?1 (productivity 23.75 h?1) obtained, represented 28-fold increase per gram cells (178 gdcw?1) when compared control (non-optimized medium), 50-fold reference ASNase production.
